They are different programming paradigms. .NET is based upon the .NET framework, a set of classes which implement many object oriented features. In other words, VB.NET is more object oriented than VB6 is. It allows for faster and easier creation of applications than VB6.
Also, support for VB.NET exists officially, and not for VB6.

.NET is more than just VB.NET....you also get support ASP.NET, ADO.NET and C#.

Will it suffice to just tell you know VB.Net? Or perhaps you could enumerate things like you know OOP, etc...?

It's always best if you mention things like OOPs, Web Services, Reflection, ADO.NET even though knowing VB.NET implies this. It lets them scan through and look at the specializations that you have.
