You would need a DevKit from Nintendo to write games for them. Could run you around 20 grand. Most likely C++, with a custom graphics API.

I would think console games would still have chunks done in ASM.
Here's a job posting by Namco - they want C and ASM, no mention of C++ even.

Nintendo reciently had tryouts for demo players. They wanted 500 people to play games at conferences and expos to demo the games and the equipment.
