Visual Studio 2005 which includes VB.NET and C#.
I don't know anything about PowerBuilder, but if you are not targeting the web, then it would not be Java.
If your background is VB, I'd say VB.NET. However, there have been raging arguments over this question repeatedly, Search this forum for .NET versus VB6 things, and you should get more results than you would want to read. They all say about the same thing, too.
It would be any version of VB.NET. Although keep in mind its not an upgrade but rather a completely new language.
Now if you are wanting to talk which language is better then thats a different story and comparison.
